What Is Mitragynine (MIT)?

Mitragynine is the primary, most abundant alkaloid found in Mitragyna speciosa leaf, commonly known as Kratom. Mitragynine makes up about 66% of Thai Kratom and around 12% of Indonesian Kratom. Therefore, Mitragynine has the most impact of Kratom alkaloids. It was Ellen Field, a medicinal chemist at the University of Edinburgh, who first isolated Mitragynine in 1921. This is pretty interesting since cannabinoids like CBD and THC found in cannabis weren’t isolated until the 1940s. This demonstrates the long history and interest in the potential of Mitragynine.

Important Notes on the Metabolism of Mitragynine

Here’s the “white lab coat” stuff if you’re interested. (There’s loads more science we could share, but we’re keeping it brief. You’re welcome.)

  • Mitragynine is an indole alkaloid found in Kratom. “Indole alkaloids” are a class of structurally similar molecules that impart various bodily activities.
  • Mitragynine interacts with the mu opioid receptors in the body, which are the same pathways attributed to some analgesic and joyful effects.
  • Mitragynine may have an inhibitory effect on the enzyme P-glycoprotein (P-gp), which underscores the importance of limiting the use of certain other drugs while taking Kratom. (This inhibitory action can slow the release of some drugs from your cells.)
  • One Kratom study demonstrated that the effects of selected indole-based Kratom alkaloids, specifically Mitragynine, “can induce antinociception* by acting in the brain”.

Antinociception definition: “the action or process of blocking the detection of a painful or injurious stimulus by sensory neurons”.

What Is 7-Hydroxymitragynine (7-OH)?

Of the total alkaloid content in the Kratom leaf, only about 2% is 7-Hydroxymitragynine in Thai Kratom and significantly less in Indonesian Kratom varieties. But don’t underestimate the power of this lesser alkaloid – it’s far more potent than the effect of Mitragynine, prompting much interest in its use. Several sources compare the effects of 7-Hydroxymitragynine to the effects of morphine, providing a quick analogy of its strength.

Though Mitragynine also interacts with the mu opioid receptors in the body, it has a lesser affinity than the highly interactive 7-Hydroxymitragynine. Even in small quantities, 7-OH can significantly impact our well-being.

The potential effects of 7-Hydroxymitragynine are similar to that of Kratom and MIT, with two exceptions:

  • 7-OH may have a more sedating effect.
  • 7-OH is highly potent, and use must be closely monitored for safety.

Important Notes on the Metabolism of 7-Hydroxymitragynine

Some more white lab coat stuff on the 7-OH effects of Kratom use:

  • 7-Hydroxymitragynine is a terpenoid indole alkaloid, a subset of indole alkaloids, with a complex (over 20 step!) biosynthesis.
  • 7-Hydroxymitragynine is an active metabolite of Mitragynine. (This means that when you ingest Kratom, the Mitragynine metabolizes into 7-OH when it interacts with certain enzymes in the body.)
  • 7-Hydroxymitragynine is a key mediator of its effects. (7-OH may “boost” the possible relieving effects of MIT.)
  • The antinociceptive effect of 7-hydroxymitragynine is likely responsible for how it can provide such intense relief. This has been demonstrated in a laboratory environment testing the effect of 7-hydroxymitragynine in mice.
  • 7-OH or (7HMG) demonstrates a greater affinity at the mu opioid receptors than Mitragynine. (This may be why 7-OH is estimated to be 14-22 times stronger than MIT.)

Key Distinctions Between Mitragynine and 7-Hydroxymitragynine

Given the different activities of Mitragynine and 7-Hydroxymitragynine, it makes sense that they’d impact us differently. While they are present in vastly different quantities in regular Kratom, nature has a way of designing things perfectly. 7-Hydroxymitragynine may be less stable in Kratom leaf, so your body naturally converts MIT into 7-OH. At least, this theory makes sense to us.

So, what are the key differences between Mitragynine and 7-Hydroxymitragynine, aside from their synthesis and in vivo mechanisms? Here are what we consider the most important differences between these two, which help outline their abuse liability and therapeutic potential:

  1. Mitragynine’s abundance in Kratom preparations is more well-known and backed by centuries of anecdotal data. (Therefore, MIT within regular Kratom and in enhanced Kratom solutions is more trusted and understood by Kratom users, which may reduce its potential for overuse or abuse.)
  2. Many Kratom users are unaware of the potency of 7-OH, and while its use may raise the therapeutic potential, this must be balanced against the risk of misusing this potent alkaloid.
  3. Both alkaloids have faced legal issues. Federally, neither Mitragynine or 7-Hydroxymitragynine are labeled as controlled substances. A few states mitigate the use of Kratom’s active alkaloids specifically. Tennessee Kratom laws take an interesting stance, only allowing the alkaloids in their natural form, outlawing any lab-adulterated compounds being marketed as “Kratom”.
  4. While both face legal battles, 7-Hydroxymitragynine has recently come under intense scrutiny as Kratom researchers question very high levels of 7-OH in Kratom products, citing it is unnatural and suggests adulteration in a lab. The AKA has also alluded to these concerns.
